The permeability and efflux of Vinca alkaloids in a Caco-2 cell model

نویسندگان

چکیده

Vinca alkaloids are used extensively in the treatment of various diseases and despite their usefulness, drug resistance, attributed to a number mechanisms associated with multidrug-resistance phenotype including overexpression P-glycoprotein (P-gp), remains serious clinical problem. To identify possible role P-gp on intestinal permeability anti-arrhythmic crude (Vingerbine) from herbaceae, Caco-2 cells were this study. The four Vingerbine constituent analyzed by high performance liquid chromatography (HPLC). Transport parameters, coefficients percent transports calculated. displayed identical tendency but dissimilar degree modulation absorptive transport direction. Vincarine Herbadine demonstrated higher-level transcellular efflux; co-presence verapamil, increased, while secretory decreased. No asymmetric permeation was observed for Herbamine Vincamajine. study suggests involvement multidrug resistance-associated proteins (MRPs) efflux Herbadine. further studies will be focused upon screening appropriate nanomedicine-based strategies combat MDR thus improve absorption alkaloids.&nbsp

Evaluation of Iron Bioavailability in Caco-2 cell Culture Model: Modification of the Original Method

Background and Objectives: In the original in vitro method for evaluation of iron bioavailability using caco-2 cell model, ferritin formation by cells is considered as a functional indicator of both absorption and utilization of iron. However, as iron is measured in the tested material and then ferritin concentration is measured in caco-2 cell lysate, it is almost impossible to have a concrete ...

Evaluation of some medicinal plants effect on Rhodamine 123 accumulation and efflux in Caco-2 cell line by flowcytometry

Background and objectives: In review of traditional Persian medicine (TPM) literature concerning multi drug therapy, a group of medicinal plants that are called "convoy drugs", agents which penetrate fast into whole or specific part of the body and accelerate delivery of drugs into specific target has been mentioned. In this study, the inhibitory effect of the aqueous extracts ...

Vinca Alkaloids

Vinca alkaloids are a subset of drugs obtained from the Madagascar periwinkle plant. They are naturally extracted from the pink periwinkle plant, Catharanthus roseus G. Don and have a hypoglycemic as well as cytotoxic effects. They have been used to treat diabetes, high blood pressure and have been used as disinfectants. The vinca alkaloids are also important for being cancer fighters. There ar...

Predicting apparent passive permeability of Caco-2 and MDCK cell-monolayers: A mechanistic model

Experimentally derived apparent permeabilities, Papp, through cell monolayers such as Caco-2 and MDCK are considered to be an in-vitro gold standard for assessing the uptake efficiency of drugs. Here, we present a mechanistic model that describes 'passive' Papp values (i.e., neglecting active transport) by accounting for the different resistances solutes encounter when permeating a cell monolay...
